I have a Phenom II X2 545 in my system and I am using a Gigabyte GA-MA770-UD3 motherboard. How would I go about unlocking the 2 locked cores on this processor to make it a quad core?

Try turning on ACC and change Bios version to Hybrid depending on your bios version.

So just turning on advanced clock calibration might unlock the cores?

I enabled ACC and switched to hybrid mode, now my motherboard is stuck on the startup screen and the hard drive activity light is solid. I don't know what the hell is wrong. I reset the cmos, but that didn't do anything. Now I'm very worried.

Nevermind, fixed it, I had to leave the battery out longer than the manual said to. I think it might have done this because I set ACC to All Cores. After I saved it said something about updating firmware. If I set it to auto will that work?

sometimes the CPU will hang with unlocked cores when ACC/hybird on stock bios,
may want to browse the net to find a beta bios for your board, it sometimes helps with stable unlocking.
